Choose CABLIVI on Day 1, in conjunction with PEX and immunosuppression
CABLIVI is indicated for the treatment of adults and adolescents of 12 years of age and older weighing at least 40 kg experiencing an episode of acquired thrombotic thrombocytopenic purpura (aTTP*), in conjunction with plasma exchange and immunosuppression1
aTTP diagnosis is based on high clinical suspicion and confirmed by ADAMTS13 testing
Early initiation of CABLIVI is recommended for acute aTTP episodes (initial and relapsing)†
A delay in CABLIVI start led to additional days of active disease
*aTTP is also known as iTTP. The terms can be used interchangeably.2
†Assumes high likelihood of aTTP (based on clinical assessment or a formal clinical risk assessment tool) and timely access to ADAMTS13 testing. In low or intermediate likelihood of aTTP, CABLIVI may be started after ADAMTS13 testing confirms a diagnosis.
ADAMTS13=a disintegrin and metalloproteinase with a thrombospondin type 1 motif, 13; ISTH=International Society on Thrombosis and Haemostasis; iTTP=iTTP=immune-mediated thrombotic thrombocytopenic purpura.

HERCULES: Study design2
A pivotal, phase 3, double-blind, randomized controlled trial of 145 patients with aTTP; patients were randomized to receive CABLIVI, PEX, and immunosuppression (n=72) or placebo, PEX, and immunosuppression (n=73).
TTP=thrombotic thrombocytopenic purpura.
HERCULES: Study endpoints2
Primary endpoint: time to confirmed platelet count response*
Key secondary endpoints
Other efficacy outcomes
PK/PD and antidrug antibodies
Safety
*Platelet count response was defined as initial platelet count ≥150 × 109/L with subsequent stop of daily PEX within 5 days.
PD=pharmacodynamic; PK=pharmacokinetic.
